چکیده

We investigated the regulatory roles of miR-141 and RICTOR genes in osteoporosis (OP) explored potential therapeutic value OP by targeting gene. The role as regulators ovariectomized (OVX) rats was determined using RT-qPCR assays models OVX rats. their expression levels related clinical features were further analyzed. functional miR-141/RICTOR pathway plasmids. lowly expressed rats, while gene highly expressed. Bioconductivity prediction analysis revealed sites for RICTOR. Promotion or inhibition mimic/ si-RNA reduced ALP activity well nuclear factor-kappa B ligand (RANKL), bone gla protein (BGP) tartrate resistant acid phosphatase (TRAP), promoting osteoprotegerin production. Healthy with elevated showed same symptoms OVX, whereas mimic injection improved Our data suggest that regulate osteoprotegerin, RANKL, BGP TRAP gene, impacting development OP.

MicroRNA-433-3p promotes osteoblast differentiation through targeting DKK1 expression

Dickkopf-1 (DKK1) is a powerful antagonist of canonical WNT signaling pathway, and is regarded as a biomarker for osteoporosis. Its expression is highly correlated with bone mass and osteoblasts maturation. In this study, mouse primary bone marrow cells and osteoblast cell lines were used. Luciferase reporter assay and western blotting methods were employed to validate if miRNA-433-3p epigeneti...

miR-628-3p regulates osteoblast differentiation by targeting RUNX2: Possible role in atrophic non-union

Atrophic non-union is a serious complication of fractures. The underlying biological mechanisms involved in its pathogenesis are not yet completely understood. MicroRNAs (miRNAs or miRs) are a type of endogenous small non-coding RNA, which participate in various physiological and pathophysiological processes. In this study, differentially expressed miRNAs were screened in patients with atrophic...

MicroRNA miR-378 Regulates Nephronectin Expression Modulating Osteoblast Differentiation by Targeting GalNT-7

MicroRNAs (miRNAs) are small fragments of single-stranded RNA containing 18-24 nucleotides, and are generated from endogenous transcripts. MicroRNAs function in post-transcriptional gene silencing by targeting the 3'-untranslated region (UTR) of mRNAs, resulting in translational repression. We have developed a system to study the role of miRNAs in cell differentiation. We have found that one of...

miR-34s inhibit osteoblast proliferation and differentiation in the mouse by targeting SATB2

A screen of microRNAs preferentially expressed in osteoblasts identified members of the miR-34 family as regulators of osteoblast proliferation and/or differentiation. Osteoblast-specific gain- and loss-of-function experiments performed in vivo revealed that miR-34b and -c affected skeletogenesis during embryonic development, as well as bone mass accrual after birth, through two complementary c...

miR-203 and miR-320 Regulate Bone Morphogenetic Protein-2-Induced Osteoblast Differentiation by Targeting Distal-Less Homeobox 5 (Dlx5)

MicroRNAs (miRNAs) are a family of small, non-coding RNAs (17-24 nucleotides), which regulate gene expression either by the degradation of the target mRNAs or inhibiting the translation of genes. Recent studies have indicated that miRNA plays an important role in regulating osteoblast differentiation. In this study, we identified miR-203 and miR-320b as important miRNAs modulating osteoblast di...
